Lipid H, also known as SM-102, is an ionizable synthetic lipid provided by Nucleosyn. This lipid can be used in combination with other lipids in the formulation of lipid nanoparticles (LNPs) for drug delivery. As an example, Lipid H-containing LNPs have been used for the delivery of mRNA-based COVID-19 vaccines. SM-102 includes a nitrogen atom which can be protonated under physiological pH to allow the LNPs to bind to the negatively charged backbone of mRNA. Tests on primates have shown that Lipid H was rapidly cleared from the body resulting in low toxicity.
