Lipid-20b
Lipid-20b is a novel ionizable lipid containing a thiophene moiety (Thio-lipid). The pH-response profile of this thio-lipid (broad ionization profile) differs from the traditional ionizable lipids. It may be related to an electron delocalization in the thiophene backbone core and its substituents, leading to a partial positive charge on the heteroatoms (nitrogen & sulfur).
Lipid-20b proved to be a robust and potent lipid for encapsulating and delivering various biologically active agents (FLuc, Cre recombinase, and EGFP mRNA) in mouse and non-human primates (NHP).
Both linoleic chains attached on the same side of the thiophene core directed Lipid-20b-containing LNPs to the liver and spleen from intravenous administration.
Lipid-20b was also suitable for the delivery of mRNA via subretinal administration in NHP with no acute toxicity and gave encouraging results in the transfection of retinal pigment epithelium and photoreceptors.
