Lipid-29d
Lipid-29d is a novel ionizable lipid containing a thiophene moiety (Thio-lipid). The pH-response profile of this thio-lipid (broad ionization profile) differs from the traditional ionizable lipids. It may be related to an electron delocalization in the thiophene backbone core and its substituents, leading to a partial positive charge on the heteroatoms (nitrogen & sulfur).
Lipid-29d proved to be a potent lipid for encapsulating and delivering various biologically active agents (FLuc, Cre recombinase, and EGFP mRNA) in mouse and non-human primates (NHP).
Both linoleic chains separated on the opposing sides of the thiophene core directed Lipid-29d-containing LNPs to the lung tissues and spleen from intravenous administration.
