Ionizable β-Sitosterol

Ionizable β-sitosterol is a newly developed sterol derivative designed for incorporation into the formulation of LNPs aimed at organ-selective mRNA delivery applications. While conventional four-component LNPs primarily deliver to the liver, the addition of ionizable β-sitosterol as a fifth component enables preferential delivery to the spleen or lungs. In SM-102-based LNPs, ionizable β-sitosterol allowed a selective lung mRNA delivery up to 78%. Similar delivery rates could be obtained with Lipid U-101-based LNPs.
