L-319 and L-322 are ionizable and biodegradable lipids. They display rapid elimination from plasma and tissues, substantially improved tolerability in preclinical studies, while maintaining in vivo potency. In vivo activity of L-319 or 322-containing LNPs was demonstrated using the mouse factor VII (FVII) gene silencing model.
