L-PGTA
L-PGTA is a new poly-ionizable lipid-modified poly(guanidine thioctic acid) polymer developed to enhance the efficiency and safety of the corresponding LNPs (G-LNPs) for mRNA cancer vaccine (G-LNPs/mRNA vaccine). L-PGTA is also biodegradable thanks to small molecules or enzymes' easy cleavage of the disulfide bridges. The G-LNPs promote the translation efficiency of loaded mRNA and decrease inflammation reactions after injection by eliminating the reactive oxygen species that inhibit translation and cause inflammation reactions. The G-LNPs/mRNA vaccine showed excellent antitumor efficacy by synergy with the immune checkpoint blockade, indicating that the L-PGTA polymeric lipid, through the G-LNPs/mRNA vaccine, may become a powerful tool in the conception of mRNA cancer vaccines.
