Lipid U-101

Lipid U-101 is a newly developed ionizable synthetic lipid designed for incorporation into the formulation of LNPs as the main component for mRNA delivery applications. While conventional ionizable lipids such as SM-102 or ALC-0315 primarily deliver to the liver, Lipid U-101-based LNPs enable preferential delivery to the spleen up to 96%. When Lipid U-101 was used in combination with Ionizable β-Sitosterol as a fifth component in the LNPs’ formulation, a remarkable lung-targeted delivery up to 71% was obtained, showing the high potency of Lipid U-101 for organ-selective delivery applications.
